Curcumin enhanced therapeutic efficacy of M. oleifera root extract and showed better antioxidant potential against beryllium toxicity.

PMID: 25853973 Abstract Title:  Co-administration of adjuvants along with Moringa oleifera attenuates beryllium-induced oxidative stress and histopathological alterations in rats. Abstract:  CONTEXT: Moringa oleifera Lam. (Moringaceae) is a rich source of antioxidants. All parts of the plant are medicinally important and have been used as traditional medicine for a variety of human ailments in India.OBJECTIVE: Therapeutic efficacy of adjuvants with M. oleifera (MO) root extract was investigated against beryllium-induced oxidative stress.MATERIALS AND METHODS: Hydroalcoholic (50% v/v) root extract of M. oleifera (150 mg/kg, p.o.) alone and combinations of M. oleifera with either piperine (2.5 mg/kg, p.o.) or curcumin (5.0 mg/kg, p.o.) daily for 1 week were administered in experimental rats against beryllium toxicity (1.0 mg/kg, i.p. daily for 5 weeks). Oxidative stress parameters including blood sugar,G-6-Pase in liver, and DNA damage were analyzed. Histopathological changes in liver and kidney were also observed.RESULTS: Beryllium enhanced lipid peroxidation (LPO), depleted reduced glutathione (GSH) and antioxidant enzymes activities, decreased blood sugar and G-6-Pase activity, and did not damage DNA. Histologically, liver was observed with structural loss and disintegration of hepatocytes, heavy vacuolation in hepatocytes, and kidney was observed with constriction of glomeruli and hypertrophy in epithelial cells of uriniferous tubules. Therapy of M. oleifera with piperine was effective; however, combination of M. oleifera with curcumin showed better therapeutic effect by reduction of LPO, elevated GSH level, maintained antioxidant enzymes activities, restored blood sugar, and G-6-Pase activity in liver together with almost normal histoarchitecture of liver and kidney.DISCUSSION AND CONCLUSION: Curcumin enhanced therapeutic efficacy of M. oleifera root extract and showed better antioxidant potential against beryllium toxicity.

Abstract:  BACKGROUND: Cardiovascular complications are the leading cause of mortality in patients with end-stage kidney disease. Research indicates that the Mediterranean diet is protective of cardiovascular disease in the general population. Components of this diet have been trialled in haemodialysis patients with the aim of reducing the risk of cardiovascular disease and improving associated risk factors. Components include fish, fruit and vegetables in the form of fish oil supplements and vitamin and antioxidant supplements. This narrative review provides an overview of observational studies, and interventional and randomised controlled trials examining the association of these supplements with cardiovascular outcomes in haemodialysis patients.METHODS: We reviewed the relevant literature by searching English-language publications in Web of Science and references from relevant articles published since 1992. Eight-seven abstracts were reviewed and 38 relevant articles were included.RESULTS: The extant literature suggests that risk of mortality is reduced in patients with a higher fish intake and those with higher serum omega-3 fatty acid levels. However, the pathways by which risk of mortality is reduced have not been fully extrapolated. While only a few studies have examined the effect of vitamin B supplementation in haemodialysis patients, these studies suggest that supplementation alone does not reduce the risk of mortality. Finally, studies examining vitamin E supplementation have drawn inconsistent conclusions regarding its pro-oxidant or antioxidant effects. Differences between studies are likely due to methodological variations in regards to dose, route of administration and treatment duration.CONCLUSIONS: Nutritional and dietary supplementation in haemodialysis patients is an area which requires larger, more methodologically robust randomised controlled trials to determine if risk of cardiovascular outcomes can be improved.

Oridonin displays potent anticancer effect against uveal melanoma cells through upregulation of Bim and inhibition of FAS.

PMID: 25545058 Abstract Title:  Oridonin induces apoptosis in uveal melanoma cells by upregulation of Bim and downregulation of Fatty Acid Synthase. Abstract:  Oridonin is an orally available drug isolated from Traditional Chinese Medicine. Previous studies with oridonin have demonstrated broad-spectrum anticancer activity in a variety of cancer types. However, the effect of oridonin in uveal melanoma has not been addressed. In this study, we aimed to investigate whether oridonin elicited anticancer activity and its underlying mechanism in human uveal melanoma cells. We demonstrated that oridonin potently reduced cell viability, induced apoptosis and inhibited clonogenic survival and growth with single digit micromolar concentrations in uveal melanoma OCM-1 and MUM2B cell lines. We found that oridonin markedly increased the expression of proapoptotic Bcl-2 family protein Bim in uveal melanoma cells, and knockdown Bim by small interfering RNA significantly attenuated oridonin-induced cell death, indicating an essential role of Bim in oridonin-mediated anticancer activity. Additionally, we observed that oridonin suppressed Fatty Acid Synthase (FAS) expression in uveal melanoma cells, and enforced FAS expression by insulin partially rescued the cells from oridonin-induced apoptosis, showing that inhibition of FAS also contributed to oridonin-mediated apoptosis. Taken together, we reported that oridonin displays potent anticancer effect against uveal melanoma cells through upregulation of Bim and inhibition of FAS. Since oridonin is a popular anticancer agent, our study therefore may have translational implication on the management of patients with uveal melanoma.

Broccoli Extract Does The "Impossible": Improves Autism Spectrum Disorder

Of all the natural substances we have reviewed with potential value in the treatment of ASD, sulforaphane, a molecule found in cruciferous vegetables such as broccoli, cabbage and Brussels sprouts, is perhaps the most promising candidate.

A groundbreaking study published in 2014 in The Proceedings of the National Academy of Sciences USA titled, "Sulforaphane treatment of autism spectrum disorder (ASD)," found that a broccoli sprout extract significantly improved the behavior of boys and men with ASD. The researchers pointed out that, presently, no conventional, mechanism-based treatment is available for ASD. Sulforaphane was selected, in part, because its physiological effects are well characterized and ideal for those with ASD:

Dietary sulforaphane, of recognized low toxicity, was selected for its capacity to reverse abnormalities that have been associated with ASD, including oxidative stress and lower antioxidant capacity, depressed glutathione synthesis, reduced mitochondrial function and oxidative phosphorylation, increased lipid peroxidation, and neuroinflammmation."

The placebo-controlled, randomized pilot study of 44 males, ages 13-27, showed that after 18 weeks of treatment with a sulforaphane-rich broccoli sprout extract, 46% had significant improvements in social interactions and 42% has significant improvements in verbal communication.

Additionally, more than half the participants showed decreases in abnormal behaviors, including irritability, hyperactivity and repetitive movements. Of note, once treatment stopped, most of the behaviors returned to pretreatment levels. The dosing schedule was determined by body weight:

  • 100 lbs or less: one capsule containing 50 µmol (232 mg) of sulforaphane-rich broccoli extract was given daily
  • 101–199 lbs, 100 µmol (two capsules of 232 mg each) of sulforaphane-rich broccoli extract was given daily
  • More than 200 lbs: 150 µmol (three capsules of 232 mg each) of sulforaphane-rich broccoli extract was given daily

Sulforaphane: A Largely Ignored Panacea-like Phytocompound

It may come as a surprise to many reading this that so simple and natural a substance as broccoli extract could have significant therapeutic value in a condition which is considered refractory to conventional treatment and whose causes, including vaccines, are considered politically incorrect to discuss, despite evidence provided by one of the CDC's senior vaccine scientists to support the connection.
